چکیده

In the prefrontal cortex, dopamine D 1 -like and M 1 muscarinic receptors are both involved in the regulation of attentional, cognitive and emotional processes but so far no information has been provided on their functional interaction. In the present study we show that in mouse medial prefrontal cortex, concomitant activation of M 1 muscarinic receptors potentiated D 1 -like receptor-induced cyclic AMP formation through a mechanism involving activation of G q/11 and the release of G protein subunits. Immunohistochemical studies indicated that the adenylyl cyclase isoforms AC2 and AC4 are expressed in mouse prefrontal cortex and that they colocalize with D 1 -like receptors with a greater association for AC4. In primary cultures of frontal cortex neurons, D 1 -like receptor-induced Ser133 phosphorylation of the transcription factor cyclic AMP-responsive element binding protein (CREB) was potentiated by concurrent stimulation of M 1 receptors. Suppression of AC4 expression with small interfering RNA transfection reduced D 1 stimulation of cyclic AMP formation and CREB phosphorylation and abolished the M 1 potentiation, whereas knockdown of AC2 had no significant Received: July 14, 2011 Accepted after revision: November 9, 2011 Published online: March 28, 2012 Prof. Pierluigi Onali Section of Biochemical Pharmacology, Department of Neuroscience University of Cagliari, Cittadella Universitaria di Monserrato IT–09042 Monserrato (Italy) Tel. +39 070 675 4321, E-Mail onali @ unica.it © 2012 S. The mitogen-activated protein kinase cascade couples PKA and PKC to cAMP response element binding protein phosphorylation in area CA1 of hippocampus.

Activation of the mitogen-activated protein kinase (MAPK) cascade recently was discovered to play an important role in synaptic plasticity in area CA1 of rat hippocampus. However, the upstream mechanisms regulating MAPK activity and the downstream effectors of MAPK in the hippocampus are uncharacterized. In the present studies we observed that hippocampal MAPK activation is regulated by both th...

In vivo imaging of CREB phosphorylation in awake-mouse brain

The cyclic adenosine monophosphate response element binding protein (CREB) is a phosphorylation-dependent transcription factor that plays important roles in memory consolidation and several neuropsychological disorders. Although analyzing the spatiotemporal pattern of CREB phosphorylation is required for elucidating the mechanism of memory consolidation, imaging of phosphorylation of a particul...

Levo-tetrahydropalmatine inhibits the acquisition of ketamine-induced conditioned place preference by regulating the expression of ERK and CREB phosphorylation in rats.

Levo-tetrahydropalmatine (l-THP) is an alkaloid purified from the Chinese herbs Corydalis and Stephania and has been used in many traditional Chinese herbal preparations for its sedative, analgesic and hypnotic properties. Previous studies demonstrated that l-THP has antagonistic activity on dopamine receptors; thus, it may have potential therapeutic effects on drug abuse. However, whether l-TH...

Cyclic AMP response element-binding protein mediates reactive oxygen species-induced c-fos expression.

Although the cyclic AMP response element-binding protein (CREB) plays an important role in the survival of neuronal cells and T lymphocytes, the role of CREB in vascular smooth muscle cells (VSMCs) is incompletely characterized. We examined the role of CREB in VSMCs stimulated with reactive oxygen species. Activation of CREB was examined by Western blot analysis with an antibody that specifical...
